Effects of ATP and Protein Syntheses Inhabitions to the Fine Structures of Small Intestinal Epithelial cells



Abstract
This study vas carried out to investigate the effects of actinomycin D (a protein synthesis inhibitor) and ethionine (a ATP synthesis inhibitor) to the fine structure of small intestinal epithelial cells, as a part of the surveys about the effects of t:he specific injurious agents to the specific cells.
Four albino rats were injected with 50 mg of ethionine per IOOmg of body weights for 8 da! inl:raperitoneally and another four rats were recieved single intramuscular injection¢¥ of 501eg actinomycin D per 100gm of body weights. Each of two rats was sacrificed at 1 and 5 days aft last injection in both groups respectively. Small intestines at the same sites were resected and observed under electron microscpe.
The results were summarized as follow:
Ethionine induced proliferation of Golgi complex and dilatation of intercellular space with accumula-tion of certain amorphous materials in the dilated space, as well as dilation of rough ER, sweIling of mitochondria and shedding and disppearance of ribosome, which could be found in the other degenerative changes. Proliferation of the Golgi complex suggested that the epithelial cells of small intestine had some degree of resistance to ethionine effects, since confromational changes from rough ER to Golgi complex required energy. Dilatation of intercellular space with accumulation of certain. material suggested that some degree of derangements in the transport mechanism.
Actinomycin D also induced characteristic nucleolar changes found in the liver cells and pancreatic acinar cells, but they were less impressive. The other changes were marked decrease of ribosome as well as degenerative changes of other organelles, which were suspected as results of direct effects of the drug to the organelles.
